AB We describe an in vitro study, within a specific ethnic group, where the
association between the HLA type and susceptibility/resistance to HIV
infection is investigated. PBMCs from healthy Bangladeshi male subjects
were infected with 4 different isolates of HIV-1 & 2 (an HIV-1 clinical
isolate, HIV-1 MN, HIV-2 CBL20, and HIV-2 ROD). Ten fold dilutions (from
10(-1) to 10(-7) of each isolate were used to infect the PBMCs. Cultures
of PBMCs were maintained for 21 days with daily inspection for
cytopathic effects and syncytia formation. Viral p24 levels were
determined on days 5, 14, and 21 after infection. PBMCs from each
subject were HLA typed by standard methods. The results obtained for the
26 male subjects studied so far showed considerable variation in the
replication of the different isolates and in the cytopathic effects
obtained. No PBMCs showed complete resistance to all 4 viral isolates
tested. However, some PBMCs showed some resistance to infection by
certain isolates as shown by an absence of both p24 core antigen and
syncytia formation during the 3 weeks in culture. No single HLA allele,
has been significantly associated with susceptibility/resistance to
infection in vitro to date. We are now performing an epitope analysis,
and the investigation is extended to include more subjects and
particularly females from the same ethnic group.
